The only way i have found to lock the car is by removing a fuse from the fuse box in the passenger side glove box.

Had this problem with my 2010 vw jetta se. Used remote and key in door to lock each time the horn would beep like the doors would lock but i was still able to open the rear driver s side door no matter what. Both front doors are fine. If the power locks seem to be working just fine from within the car then you need to replace your key fob battery.
